Fluorescence microscopes quantify the structure and biochemistry in specimens with submicron spatial resolution based on fluorescence contrast. This article covers the spectroscopic principles underlying fluorescence imaging. The design considerations for several basic forms of fluorescence microscopes will be presented. Fluorescent probes are classified and their optical properties will be discussed in the context of their uses in microscopic imaging. Finally, advanced modalities for functional imaging and their biomedical applications are described.
